Item type:Publication, Comparison of ELISA and chemiluminescence immunoassay methods for quantification of human Placental growth factor in serum(Croatian Society of Biologists in Health Care, 2020-06-01); ; ;Bashkim Ismaili; Placental growth factor (PlGF) is crucial during placental development in early pregnancy. Several studies in pregnancies with complications such as preeclampsia or small for gestational age neonates find that PlGF levels are significantly lower in the first trimester, which implies that the concentration of PlGF could be used as an early screening biomarker for these conditions. This study aimed to compare the performance of chemiluminescence immunoassay (CLIA) and enzyme-linked immunosorbent assay (ELISA) for the quantification of human PlGF in serum. This is a comparative study on 88 pregnant women in the first trimester subjected to measurement of PlGF in serum using two commercially available kits: Human PlGF Quantikine HS ELISA (R&D Systems) and PlGF CLIA (Snibe). The overall coefficient of correlation between the tests was 0.93. When the cut-off value of 40 pg/mL was applied, it dropped significantly to 0.50 towards the lower values, while remaining an excellent 0.91 in the group with higher concentrations of PlGF. While R&D Systems’s ELISA seems to have better sensitivity, it is not very convenient to use for a small number of samples. Snibe’s CLIA automated method is user-friendly, fast and powerful. Both tests show excellent performance when indicating risk-free pregnancies. - Some of the metrics are blocked by yourconsent settings
Item type:Publication, Comparison between the homologous BNT162b2 and the heterologous Gam-COVID-Vac/BNT162b2 vaccine regimen in republic of North Macedonia(Croatian Society of Biologists in Health Care, 2022-12-16) ;Nedelkoski, Stefan; ; ; <jats:p>The medical and socio-economic consequences that stemmed from the COVID-19 pandemic, forced the healthcare policymakers in Republic of North Macedonia to rely on five different vaccines against the SARS-CoV-2 virus, in order to reach a satisfactory level of herd immunity. It is here where we got the idea to compare the heterologous Gam-COVID-Vac/BNT162b2 regimen to the homologous BNT162b2 regimen, with our main focus being the immunogenicity differences between the two of them. Additionally, we researched the variation in humoral immune response relative to age strata; the reactogenicity differences; and discrepancies in SARS-CoV-2 infection incidence between the two regimens. To achieve this, antibody titers in sera samples from fifty-three (53) healthcare workers, divided in heterologous and homologous group, were analysed at six different time checkpoints. Our results showed robust immunogenic response after the administration of the booster dose (4. 2-fold increase in antibody titers), followed by a slower-waning humoral immune protection in the heterologous regimen, compared to the homologous BNT162b2 schedule, furthermore confirmed by non-inferiority testing (Geometric Mean Ratio=0,98) at the final checkpoint. That, coupled with the similar reactogenicity (p=0,767) of both regimens, imply that the Gam-COVID-Vac/BNT162b2 combination might be a feasible approach in the effort to contain the COVID-19 pandemic.</jats:p> - Some of the metrics are blocked by yourconsent settings

Item type:Publication, HLA profile of the Macedonian, Albanian and Macedonian Muslim donors in the Macedonian Bone Marrow Donor Registry(Elsevier BV, 2019-09); ; ; ;Bojan DobrevskiThe aim of this study was to determine the HLA allele and haplotype frequencies of the volunteer donors from the Macedonian Bone Marrow Donor Registry (MBMDR). We analyzed 1541 donors, from different nationalities and presented the HLA allele and haplotype frequencies for Macedonian, Albanian and Macedonian Muslims, most numerous nationalities in MBMDR. Difference between the three groups was observed for allele frequencies in HLA-C and HLA-DRB1 loci. The most common haplotype in Macedonian was HLA-A*01-B*08-C*07-DRB1*03, while in Albanian and Macedonian Muslims HLA-A*02-B*18-C*07-DRB1*11. Item type:Publication, Incidence of β-lactoglobulin intolerance among lactose intolerance suspected population and diagnostic approach(Macedonian Pharmaceutical Association, Ss. Cyril and Methodius University in Skopje, Faculty of Pharmacy, 2022-12-31); ; ;Zhivikj, Zoran; Milk and dairy products have high nutritional value due to its protein, vitamin, mineral and fatty acid content (FAO, 2013). However, regular usage of these products in some individuals may result in manifestation of adverse reactions, such as lactose intolerance (LI) and cow`s milk protein intolerance (CMPI). LI is a syndrome with primarily gastrointestinal symptoms as a result of lactase deficiency in the intestinal mucosa and lactose malabsorption. It can be genetically driven, known as primary LI, where homozygous LCT-13910C and LCT-22018G variants are coding low lactase activity, or secondary LI due to damaged mucosa. CMPI is an immune mediated reaction to milk`s proteins causing abdominal pain, diarrhea, vomitus, nausea etc. Since LI is highly prevalent in the population, while clinical features of LI and CMPI are similar, the last is often misdiagnosed. Treatment of both conditions is mainly based on food restrictions excluding lactose or cow`s milk proteins, so relevant diagnosis is important in successful disease management (Di Constanzo et al., 2019). Item type:Publication, DETECTION OF A RARE MUTATION IN A NOONAN SYNDROME SUSPECTED PATIENT: A CASE REPORT(Croatian Society of Biologists in Health Care (CROBIH), 2020-04); ; ; ;Milanovski GorjanNoonan syndrome (NS) is a genetic autosomal dominant condition, caused by mutations in PTPN11 and other genes. The aim of this report is to highlight a finding of a rare mutation in the RAF1 gene in a six-year-old child evaluated for Noonan Syndrome. An Ampliseq Research Panel covering A2ML1, BRAF, CBL, HRAS, KRAS, MAP2K1, MAP2K2, NRAS, PTPN11, RAF1, RIT1, SHOC2, SOS1 and SPRED1 genes was used on the Ion Torrent platform. Out of 54 variants detected, a single nucleotide missense mutation c.483T>G in the RAF1 gene was classified as likely pathogenic, based on a single previous submission to Clinvar. Further investigations may shed light on the possible role of this variant in the pathogenesis of Noonan Syndrome and other RASopathies. - Some of the metrics are blocked by yourconsent settings
